Papain catalysed peptide synthesis was followed to determine the optimum conditions for adsorption and covalent binding immobilization methods. The synthesis of the dipeptide Gly-Phe was studied in two different reaction systems: a) For adsorbed papain, in an organic medium containing low water concentration, b) For covalently bound enzyme, in a two-liquid phase system, using trichloroethylene as organic phase.
