The present project aims to develop a combined strategy for the repair of traumatic spinal cord injuries, based in the combined application of neuroprotective drugs and cell transplants, that will improve significantly the results of single treatments used in spinal cord injured models. The combined drugs-cell treatment proposed is based on three branches to promote firs neuroprotection, by administering potent drugs within an initial time window, then axonal regeneration of lesioned spinal tracts and also restitution of the lost motoneurons. The latter two points will be achieved by the combined transplants of ensheathing glia cells and embryonic stem cells, previously pre-differentiated in vitro. The former will act to exert protective action and in addition to stimulate and guide axonal regeneration and to promote a local ambiance permissive for neuronal differentiation of the stem cells in vivo. The proposed project will be initially developed in vitro by using a model of neurotrauma, based on organotypic spinal cord cultures. In parallel, the stem cells will be prepared by genetical modifications to facilitate their follow-up and identification in vivo, after transplantation. In the following phase each developed strategy will be evaluated in vivo after partial lesion by contusion or complete lesion by section of the spinal cord in adult rats. Finally, the combinations of pharmaco-cellular therapies yielding better results in the previous steps will be assessed in the same animal models to evaluate neuroprotection, neurogenesis and regeneration by means of electrophysiological, functional and histological techniques (...).
