This research project proposes the use of the rhodopsin and bacteriorhodopsin systems as models for the G protein-linked membrane receptors. Emphasys will be given to the structural aspects that can be significant for this receptor family. The most noticeable ones are the characteristics of the ligand binding site (the retinal environment in rhodopsin and bacteriorhodopsin) the mechanism of signal transmission from the binding site to the cytoplasmic loops, and the characteristics of the G protein binding site. These aspects will be mainly studied by means of high-resolution Fourier transform-infrared spectroscopy in order to monitor the structure of several modified forms of both proteins (specific cleavage on some loops and the C-terminal segment and modification of structural constants present in the receptors family), as well as using fluorescence methods to quantify the functionalit
