Description
The long-term objective of this project is to understand theinteraction of the tetanus toxin (TeTox) with the neuronal membranereceptor(s) and the molecular basis for neurotoxicity resulting fromthis interaction. For this, we have recently described that TeToxproduces a marked intracellular redistribution and down-regulation ofCa"super2"+/phosphatidylserine-dependent C kinase protein (CKP)activity. Our finding indicates that CKP activation and serotoninaccumulation may be closely related. These discoveries justify adetailed characterization of the neuronal membrane receptors. The specific aims will include a) isolation and characterization oftoxin-receptors in nerve-cell culture, primary neurons and membranepreparations; b) effects of the TeTox towards these receptors ontransmitter release; c) effects of the TeTox on the inositolturnover, d) changes on protein phosphorilation after "in v
