Genetic and biochemical determinants of alcohol toxicity and addiction: the role of Alcohol dehydrogenase isozymes

Alcoholism is the most widely spread drug addiction and dependence in Europe, and it is now clear that genetic factors are involved in its development. Ethanol oxidation is mainly catalyzed by liver alcohol dehydrogenase (ADH). The objective is the search in the Europen population for specific ADH isozyme patterns and activities which could represent higher susceptibility to alcoholism and to ethanol related diseases. The study will focus on the liver enzyme, and on stomach ADH, which may play a role in the elimination of ingested ethanol (first-pass metabolism). An additional objective is the characterization of the gastric "sigma"-ADH isozyme. Liver and stomach biopsies and blood samples from patients will be collected in Barcelona, Bordeaux, Heidelberg and Stockholm, and used for ADH activity, isozyme patterns, genotyping studies.
