The aim of this project is to elucidate the role of attentional processes as an essential step in cognitive tasks. The main objectives are: 1) to set up and to do the pharmacological validation of several attentional and working memory behavioral tasks, 2) to study the effects of environmental enrichment on these models, on the immediate early gens activity and in parameters of neurochemical modulation related to the cholinergic system. Male Sprague-Dawley rats will be used as experimental animals. In order to obtain behavioral parameters for stimulus discrimination, reaction time o selective attention, we will use attentional and working memory tasks such as the object recognition tests, the prepulse inhibition, the nine hole box, the discriminative learning and the Atlantis platform in the Morris Water Maze. The pharmacological validation of these tests will be done by using acetylcholinesterase inhibitors, muscarinic or nicotinic agonists and antagonists. Other experiments will be done to study the effects of environmental enrichment (administered during 8 weeks in the infantile-juvenile period) in those tasks. One hour after the tests, the animals will be sacrified and their brains used to study the activity of immediate early genes such as tPA and NGFI-A. Other brains will be used for autoradiography studies to determinate the neurochemical modulation of the cholinergic system induced by the environmental enrichment. Experimental designs will consist in dose-response and factorilas (in those experiments with environmental enrichment) and they will be uni or multivariant statistically analysed. From the obtained results, we could get new approaches to prevention and treatment of cognitive disfunctions and impairment
