The main objective of this study is the design of a new, optimized extender for medium-to-long storage (until 15 days) of refrigerated porcine semen. This extender would allow to maintain the full fertilizing potential of the stored semen sample. The approach will be found in a previous deeper knowledge of the mechanisms by which boar spermatozoa are optimizing their energy status. This knowledge will allow us to design an extender which maintained an optimal energy environment for maintaining the boar sperm basal energy metabolism as long as possible. To perform this approach, we will start assuming that the main energy source of mammalian spermatozoa are monosaccharids. First of all, we will study the boar sperm metabolism when the cells were incubated in the presence of various monosaccharides, such as glucose, fructose or sorbitol. This will indicate which monosaccharide is consumed by boar spermatozoa rendering a better net energy output. This will also indicate which sugar will be the best in order to a long term maintainement of boar sperm functionality. Next, we will design several extenders from the most efficient sugar. We will determine the ability of boar sperm to maintain their "in vitro" function in these extenders. Finally, we will study the results obtained after insemination of sows with the two extenders which had had the best "in vitro" results. This will allow to comprove the real utility of the new designed, optimized, extenders.
