The complement system in fish is a key mechanism in the activation of the immune response repertoire. This is due to the special characteristics of its central protein C3: ability to link to several surface cells and to react with a number of proteins, and specially its variability. Knowing how and when C3 is activated and its quantification may result in a significant tool for fish immune therapy. Thus, the aims of the present project are: - To assess which is the mechanism by which C3 activates macrophage cells - To develop an ELISA test for C3 isoforms to determine the activation of such isoforms and quantify their levels. - To quantify their activation after subjecting fish to several in vivo experimental conditions (occurrence of micro-organisms and the influence of stress conditions and diet deficiency). The previous characterisation of C3 proteins in sea bream, the availability of techniques for studying membrane channels and the collaboration of Univ. of Pennsylvania will enable the success of the project. As a final result it is envisaged to determine the mechanism of macrophage activation by C3, and to obtain a test for assessing the degree of immune activation based in the complement system. This will allow to know whether immunoestimulation has occurred after different environmental or culture conditions in sea bream. This study may lead to further work on new vaccines based on the activation of complement.
