Description
Human liver has several isozymes of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) that are involved in ethanol metabolism. Chronic alcoholism may cause liver injuries of different severity that appear to be related to decreased ADH and ALDH activities. Classically, two kinds of approaches have been taken to better understand the relationship between the enzymology of ethanol metabolism and its toxic effects. Most of the studies on enzymology were done "in vitro" at conditions far from being physiological, while the studies on toxicity were undertaken in rats, with the limitations of using an animal model. The main goal of the present proposal is to establish hepatic-cell cultures as an integrative model for research on enzymology and toxicity of ethanol.
