In our laboratory we have previously that the plasmid pKM101 contains some genes, differents of "mucAB", that give resistance to "E. coli" in the treatment with 4-quinolones (4-Qs), such as nalidixic acid or ciprofloxacin. The main objectives of this proposal are: a) the cloning and sequencing of this (or these) gene(s); b) the study of their expression and regulation; c) to determine if this mechanism of resistance is particular for inhibitors of the DNA gyrase (as 4-Qs) or could be involved in the resistence to other chemicals that provoke other kind of damages in the DNA. To achieve the mentioned objectives, different regions of the pKM101 will subcloned and we will determine which of these regions contains the gene(s) involved in the resistance to 4-Qs by an easy method developed by us.
