Resum

The development of novel and sensitive strategies for DNA hybridization and detection has become increasingly important in different fields, such as food quality and safety control in food industry. For this purpose, enzyme markers can be used as sensitive electrochemical reporters for DNA hybridization. A sensitive, selective and rapid genomagnetic assay -based on the enzymatic labelling with electrochemical detection of DNA- by using a novel magneto electrode has been developed. The DNA target is selectively bound by its hybridization with the biotinylated capture probe on streptavidin magnetic beads, and then separated from its biological complex matrix by using the magneto electrode. The sensitive DNA electrochemical detection is finally achieved by the enzyme marker HRP. The novel methodology is also shown to be useful for the rapid and sensitive detection of Salmonella spp. The main features of this approach are compared with classical DNA analysis.
